Mouse Lactate dehydrogenase-A (LDH-A)

 

Product Description: Vertebrate lactate dehydrogenase (LDH) is an oxidoreductase which catalyzes the interconversion of L-lactate and pyruvate. It consists of 4 subunits which may be of 2 different types: M (muscle) and H (heart), encoded by the LDH-A and -B genes, respectively. Five different isoenzymes are therefore possible, depending on the subunit composition. A third isoform, LDH-C or LDH-X, is expressed only in the testis. The LDH-A gene is located on chromosome 11p15.4. Multiple transcript variants encoding different isoforms have been found for this gene. Mutations in LDH-A have been linked to exertional myoglobinuria, which occurs as a result of vigorous exercise under hot and humid conditions. Interestingly, LDH-A has been found to interact via its NAD-binding domain with the AU-rich elements of many mRNAs, implicating its gene regulatory function. The recombinant mouse LDH-A is expressed as a GST-fusion protein. The fusion junction of the protein harbors an engineered thrombin cleavage site and a protein kinase A (PKA) phosphorylation site. The GST portion of the fusion protein can be cleaved off by thrombin if necessary. The fusion protein can be radioactively labeled with 32P-r-ATP and PKA. The product is for research use only.
