Glutathione S-Transferase (GST) Assay Kit

10x Cell Lysis Solution: 25 ml, store at 4ºC
50x GST Buffer: 2 ml, store at 4°C (for 500 assays)
200x CDNB: 0.5 ml, store at -20°C
GSH: 0.2 g, store at 4ºC

Product Description: The glutathione S-transferase (GST) family comprises multiple classes of abundant enzymes with highly specific and
overlapping functions. More than 20 GST class members have been identified in Homo sapiens. GSTs are well known for their ability to catalyze
the conjugation of reduced glutathione (GSH) to electrophiles on many substrates for the purpose of detoxification. GSTs also catalyze GSH-
dependent isomerization reactions involved in the synthesis of several prostaglandins and steroid hormones. In addition, GSTs have been
implicated in cell signaling, oxidative stress, cancer development, neuronal degeneration and chemotherapeutic drug resistance. Most GSTs
interact with the compound 1-chloro-2,4-dinitrobenzene (CDNB), forming the basis for a spectrophotometric assay for GST activity (Habig et al.,
PNAS 71:3879-3882, 1974). The GST activity assay kit is based on the use of CDNB, and can be employed to measure the activity of a purified
GST enzyme or total GST activity of a crude cell/tissue extract. GSH conjugation to CDNB causes an increase in the absorbance at 340 nm. The
assay requires a 0.2-ml Quartz cuvette or UV-transparent 96-well plate for measuring kinetic changes in O.D.340 nm. Each assay kit is for 500
assays.
