Product Description: Glycerol-3-phosphate dehydrogenase (GPDH) catalyzes the reversible conversion of dihydroxyacetone phosphate (DHAP) to glycerol-3-phosphate (G3P). In the glycerol phosphate shuttle, DHAP is reduced by the cytosolic GPDH (GPDH1 or cGPDH), which is NADH dependent, to G3P. G3P can diffuse from the cytoplasm to the inner mitochondrial membrane where it is oxidized back to DHAP by the mitochondrial GPDH (GPDH2 or mGPDH), which is FAD-dependent. The cytosolic and mitochondrial GPDH enzymes thus constitute an electron shuttle mechanism that facilitates the transfer of reducing equivalents from the cytosol to mitochondria. Since G3P also plays a role in lipid biosynthesis, GPDH serves to coordinate carbohydrate and lipid metabolism. GPDH activity is also known to increase during adipogenic differentiation. The enzyme activity assay kit can be used to quickly assess potential changes in the expression and regulation of GPDH1. The non-radioactive colorimetric GPDH assay is based on the reduction of the tetrazolium salt INT in a NADH-coupled enzymatic reaction to formazan, which is water-soluble and exhibits an absorption maximum at 492 nm. The intensity of the red color formed is increased in the presence of increased GPDH activity. The kit is stable for at least one year if handled and stored properly.
